You can't have started Botanics Studies quest because you won't be able to start this one.

Way to Island

In order to go to Twin Rocks Isle, you have to go to gym in Mossdeep.
Use Twin Rocks Ticket and talk with NPC Tate and Liza.

Path to quest npc

You will find there NPC Jonnes at which you start mission.

Mission 1

Npc gives you mission to examine the temperature for all 34 pokemons, that are located on this island in the ice part.
Use thermometer on pokemon.

Pokemon list:

5. Piloswine. 6. Nincada. 7. Poliwag. 8. Dratini.
9. Wingull. 10. Swinub. 11. Smoochum. 12. Vanillite.
13. Lapras. 14. Cubchoo. 15. Spheal. 16. Sealeo.
17. Ponyta. 18. Snorunt. 19. Vulpix. 20. Swablu.
21. Seel. 22. Sentret. 23. Snubbull. 24. Omanyte.
25. Amaura. 26. Bagon. 27. Surskit. 28. Poliwhirl.
29. Jynx. 30. Piplup. 31. Furret. 32. Butterfree.
33. Pelipper. 34. Horsea.

You can use thermometer on yourself to see which pokemons you are missing.
Once you check temperature of all pokemons go back to npc and finish mission.

Mission 2

In this mission you will have to kill requested ammount of pokemons from these types:
Water: 150
Dragon: 150
Fairy: 50
Fire: 150
Ground: 100
Ice: 400
Once you kill all of them go back to npc.

Mission 3

Now npc tells you to kill these 6 bosses: Flamehorse, Aquadra, Appa, Clouddy, Madog and Iceless Glaceon. Rooms with bosses are scattered around ice part of island. One of these rooms have lever at the middle of it and this one leave as a final one. When you kill all bosses go back to NPC and end mission.

Reward

• Sleeping Glaceon plush,
• Possibility to evolve Eevee into Glaceon.
